ABSTRACT

Objective To investigate the protective effect of Shenxi Dan,a recipe for cooling blood,activating blood and inducing resuscitation,on multiple organ damage of rats with endotoxic shock.Methods The SD rats were randomly divided into normal group,model group and Shenxi Dan group.Except for the normal group,the rats in the other groups were given intraperitoneal injection of D-galactosamine (GalN) and intravenous injection of lipopolysaccharide (LPS) to induce endotoxic shock.The changes in mean arterial pressure (MAP) during the entire process were monitored.Serum samples and liver and lung tissues were collected after infusion for 0 h,2 h,6 h,respectively,and then the amount of whole blood cells,serum contents of total bilirubin (TB),lactate dehydrogenase (LDH),aspartate aminotransferase (AST),and alanine aminotransferase (ALT) were detected.Meanwhile,the pathological changes in lung and liver tissues after hematoxylin-eosin (HE) staining were observed under light microscope.Results Compared with the model group,the decrease degree of the blood pressure was mild,platelet count(plt) was not decreased obviously,while the increase of serum TB,LDH,AST,ALT contents was significantly inferior,and inflammatory pathological changes of lung and liver tissues significantly were much relieved in Shenxi Dan group.Conclusion Shenxi Dan can improve lung and liver function of endotoxic shock rats,reduce the pathological damage,and improve microcirculation,which is helpful to clinical treatment of endotoxic shock.

ABSTRACT

Objective: To obtain the optimum prescription of the rhubarb lactobacillin dispersible tablets.Methods: The kind and amount of the excipient were investigated by using the disintegrating time to establish the optimum prescription and preparation process.Results: The tablets were prepared by compressing moist granulation with the PVPP and CMS-Na as the disintegrant,MCC as diluent,sodium alginate as sweller,magnesium stearate as lubricant and 80% alcohol as adhesive agent.The preparation complied with the requirement of Pharmacopeia of the People's Republic of China(2005edition).Conclusion: This prescription of the tablet is reasonable and the preparation process is feasible.

ABSTRACT

OBJECTIVE: To investigate the effects of Yinchen Shufu Decoction on hepatocyte apoptosis and the expression of its apoptosis-regulating gene Bcl-2 and Bax in Yin-jaundice rats. METHODS: Forty-eight rats were divided randomly into 4 groups: the normal control group, Yin-jaundice model group, Yang-jaundice model group, Yinchen Shufu Decoction treatment group. The TUNEL assay and the immunohistochemistry assay were used to detect the apoptosis of hepatocytes and the expression of Bcl-2 and Bax in hepatocytes respectively. RESULTS: The rate of apoptosis cells in Yin-jaundice model group was higher significantly than that in Yang-jaundice model group and normal control group (P<0.01), the expression of Bcl-2 in Yinchen Shufu Decoction treatment group was higher significantly than that in Yin-jaundice model group (P<0.01 or P<0.05), and the expression of Bax in it was lower significantly than that in Ying-jaundice model group (P<0.01 or P<0.05). CONCLUSION: Yinchen Shufu Decoction can prevent hepatocyte apoptosis perhaps by up-regulating the expression of Bcl-2 and down-regulating the expression of Bax. It is one of the mechanisms of its treatment on Yin-jaundice.

ABSTRACT

Objective To investigate the effect and mechanism of Artemisia Argyi extracting solution on anti-hepatic fibrosis.Methods Twenty-four experimental Wistar rats were randomized into four groups:Artemisia group,Danshen group,physiological saline group,and blank control group,with 6 in each.The Artemisia group was treated with 20% Artemisia Argyi extracting solution,the Danshen group with Danshen (salvia) solution,and the physiological saline group with physiological saline by gavage,20ml/kg.After treating three times per day for 3 days,sera were obtained and made a mixture with DMEM to be volume fraction as 5%,10%,and 20%.Effect of different volume fraction of extracting solution on TGF-?1 Smad3 and Smad7 mRNA and protein were detected.Results After treating with 5%,10% and 20% Artemisia Argyi extracting solution contained rat serum for 24h,contrasted to the black control group,the expression of TGF-?1 mRNA level was (0.82?0.03),(0.72?0.03) and (0.67?0.04) times,and the expression of TGF-?1 protein was (0.66?0.09),(0.34?0.05) and (0.31?0.07) times respectively;contrasted to the physiological saline group,the expressions of TGF-?1 mRNA and protein were significantly decreased (P

ABSTRACT

The results of this study revealed that there was less discoid cell and more pseudopodium, more thrombocytes tended to agglutinate with increased proportion of hyper - glycogenic platelets. There were also platelets sticking to RBC. WBC and lymphocytes. The glanules within the platelates were decreased or even fused together, unable to differentiate them. The vascular system were dilated and proliferated. Abnormal granules were seen with its size similar to a - granules, covered with membrane, with cytoplasm and metabolites inside. Under the revulsion of ADP, there was 70. 28% agglutining body. Most intercellular spaces revealed unclear bridge - form connection or even disappeared with fusion of cellular membrane. All these changes were more distinctive than that in the control group, demonstrating the changes in ultrasturcture and agglutination function might be one of the causes of blood stasis in hepatopathy.
